I don't think it's anything but an interesting coincidence. I think when Stoker was researching "Dracula", the birthday may have struck him as interesting or amusing, but I don't think it unduly influenced him. His interest mainly lay in Dracula's name, his links with Transylvania, and his bloodthirsty reputation.
